干细胞因子(SCF)是一种早期作用的细胞因子,在胚胎和成人造血过程中起着关键作用。SCF促进细胞存活、增殖、分化、粘附以及在造血系统多个层面的功能性激活。SCF通常与其他细胞因子如血小板生成素和Flt3/Flk-2配体一起用于促进培养的原始造血干细胞和多能祖细胞的扩增(Martin等;Kent等)。SCF与多种生长因子协同作用,包括IL-2、IL-3、IL-6、IL-7、G-CSF和促红细胞生成素,可促进髓系和红系祖细胞以及淋巴系祖细胞的增殖和分化(Broudy)。SCF也是肥大细胞和嗜酸性粒细胞的主要生长和激活因子。


SCF以两种生物活性剪接形式存在:可溶性形式和跨膜异构体。与其受体(c-Kit酪氨酸激酶受体;CD117)结合后,SCF可激活PI3K, JAK/STAT和MAPK通路。据报道,SCF和c-Kit信号转导在色素沉着、生育力、血管生成、通过c-Kit阳性的Cajal间质细胞促进肠道蠕动以及神经干细胞和祖细胞向脑损伤部位迁移中发挥重要作用。本产品不含动物成分。

(A) The biological activity of Human Recombinant SCF, ACF was tested by its ability to promote the proliferation of TF-1 cells. Cell proliferation was measured after 72 hours of culture using a fluorometric assay method. The EC50 is defined as the effective concentration of the cytokine at which cell proliferation is at 50% of maximum. The EC50 in the example above is 1 ng/mL.
(B) 1 μg of Human Recombinant SCF, ACF was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining.
