概述

白细胞介素3 (IL-3)是一种物种特异性的多效性细胞因子,可促进多能造血干细胞和谱系特异性祖细胞的存活和增殖,并促进其分化为大多数谱系的成熟细胞,包括肥大细胞、嗜碱性细胞、中性粒细胞、嗜酸性细胞、巨噬细胞、树突状细胞、红细胞和巨核细胞(Fung等;Metcalf等人;Broughton等人)。IL-3由活化的T细胞产生,通过促进肥大细胞、嗜碱性粒细胞和嗜酸性粒细胞分泌组胺、IL-4和IL-6等炎症介质,在炎症和过敏中具有生理作用。小鼠IL-3受体由一个独特的α亚基(CD123)和两个β亚基组成,一个特异于IL-3 (βIL-3),另一个与IL-5和GM-CSF受体(β公共链,βc或CD131)共享。IL-3与含有α亚基和α亚基之一的异二聚体受体结合可激活JAK/STAT、MAPK和PI3K信号通路(Scott & Begley)。

(A) The biological activity of Mouse Recombinant IL-3 was tested by its ability to promote the proliferation of Ba/F3 cells. Cell proliferation was measured using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum. The EC50 in the above example is 2 - 3 pg/mL.
(B) 1 μg of Mouse Recombinant IL-3 was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Mouse Recombinant IL-3 has a predicted molecular mass of 15.2 kDa.
