干细胞因子 (SCF) 是一种早期作用的细胞因子,在胚胎和成人造血的调控中起着关键作用。SCF 促进细胞存活、增殖、分化、粘附以及在造血系统多个层面的功能性激活。SCF 与其他细胞因子,例如血小板生成素和 Flt3/Flk-2 配体一起,常用于促进培养的原始造血干细胞和多能祖细胞的扩增 (Martin et al.; Kent et al.)。SCF 与多种生长因子,包括 IL-2、IL-3、IL-6、IL-7、G-CSF 和促红细胞生成素协同作用,促进髓系和红系祖细胞以及部分淋巴系祖细胞的增殖和分化 (Broudy)。SCF 也是肥大细胞和嗜酸性粒细胞的主要生长和活化因子。

SCF 有两种生物活性剪接变体:可溶性和跨膜型。与其受体(c-Kit 酪氨酸激酶受体;CD117)结合后,SCF 可激活 PI3K、JAK/STAT 和 MAPK 通路。据报道,SCF及其介导的c-Kit信号在多个生理过程中发挥重要作用,包括色素生成、生育力、血管生成、Cajal间质细胞介导的胃肠道运动调节,以及神经干/祖细胞向脑损伤区域迁移等过程。

(A) The biological activity of Human Recombinant SCF (P. pastoris-expressed) was tested by its ability to promote the stimulation of human TF-1 cells. The EC50 is defined as the effective concentration of the growth factor at which stimulation is at 50% of maximum. The EC50 in the above example is ≤ 2 ng/mL. (B) 2 μg Human Recombinant SCF (P. pastoris-expressed) was resolved with SDS-PAGE under non-reducing (-) and reducing (+) conditions. Human Recombinant SCF has a predicted molecular mass of 18.6 kDa.
