表皮生长因子受体(EGFR)是一种 I 型跨膜蛋白和酪氨酸激酶受体。EGFR已被证实可与表皮生长因子(EGF)家族的某些配体结合,包括EGF、amphiregulin、TGF-α、betacellulin、epiregulin、肝素结合型EGF以及neuregulin-2α。EGFR配体结合会诱导EGFR的同源二聚化,以及EGFR与ErbB2或与配体激活的ErbB3或ErbB4的异源二聚化(Schlessinger)。二聚化可导致激酶激活、磷酸化和细胞信号传导,主要通过MEK/ERF和AKT通路介导(Navlonic等)。EGFR信号传导已被证实可调节细胞增殖、分化、迁移及凋亡。EGFR水平升高与致癌作用相关(Maihle等)。蛋白质在多肽链的羧基端含有一个 His 残基标签。

(A) The biological activity of Human Recombinant EGFR was tested by its ability to promote the proliferation of BALB/c 3T3 cells in the presence of 25 pg/mL EGF. Cell proliferation was measured using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum. The EC50 in the above example is 0.81 µg/mL. (B) 2 μg of Human Recombinant EGFR was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Human Recombinant EGFR has a predicted molecular mass of 69.4 kDa.
