概述

10C2抗体与人巢蛋白反应,巢蛋白是一种由发育中的神经系统中的胚胎神经上皮干细胞、胰岛细胞和间充质祖细胞表达的VI类中间丝蛋白。在胶质母细胞瘤和许多原发性和转移性黑色素瘤中也观察到Nestin的表达,它是评估肿瘤内皮细胞新生血管的有用血管生成标志物。Nestin是神经祖细胞存活和更新所必需的,并通过与vimentin或α -间连蛋白形成异源二聚体在细胞重塑中发挥作用,这一过程似乎受到磷酸化的严格调节。巢蛋白在分化神经元轴突伸长过程中定位于生长锥,可能在生长锥引导中起作用。Nestin的表达在分化过程中下调,通常在胚胎期E18消失。据报道,10C2抗体与猴巢蛋白有弱交叉反应,而与啮齿动物巢蛋白无交叉反应。在western blots上观察到~220 - 250kda的双峰带。

(A) Human neural progenitor cells (NPCs) were generated from induced pluripotent stem (iPS) cells using STEMdiff™ Neural Induction Medium and cultured on Corning® Matrigel®-coated glass coverslips. Cells were fixed and labeled with Anti-Human Nestin Antibody, Clone 10C2, followed by donkey anti-mouse IgG, Alexa Fluor® 488. Inset shows cells incubated with a mouse IgG1, kappa isotype control antibody followed by donkey anti-mouse IgG, Alexa Fluor® 488.
