RPA-T4抗体可特异性识别CD4。CD4是一种分子量约为59 kDa的单链1型跨膜糖蛋白,属于免疫球蛋白 (Ig) 超级家族。CD4包含四个胞外免疫球蛋白样结构域 (D1-D4)。RPA-T4识别该蛋白的D1结构域,其结构类似于免疫球蛋白可变结构域。CD4在大多数胸腺细胞和T细胞亚群(辅助性T细胞)中表达水平相对较高,而在外周血单个核细胞和巨噬细胞中表达水平较低。CD4与MHC II的非多态性区域结合,并通过增强TCR和MHC II抗原复合物之间的亲和力,在MHC II限制性抗原识别中作为T细胞受体(TCR)的共受体。CD4还通过胞内结构域与细胞质酪氨酸激酶(如Lck)的相互作用,放大从TCR到细胞质的信号。此外,CD4是人类免疫缺陷病毒(HIV)的受体。RPA-T4抗体的结合可阻断HIV结合和混合淋巴细胞反应(MLR),并通过干扰CD4与HLA-DR的结合来抑制体外CD4+ T细胞的激活。它也能部分阻断SK3抗体的结合。研究发现,RPA-T4抗体的表位对甲醛固定敏感。
