白血病抑制因子
白血病抑制因子 (LIF) 是一种白细胞介素 6 类细胞因子,可调节多种发育功能。LIF 与 LIF 受体 (LIFR) 结合后,会与 gp130 结合并激活 JAK/STAT 和 MAPK 信号传导 (Auernhammer & Melmed; Suman et al.)。LIFR 激活 STAT3 对于维持小鼠胚胎干细胞表型至关重要 (Niwa et al.)。LIF 由子宫内膜产生,通过调节胚泡附着后滋养层细胞的增殖、侵袭和分化,在着床过程中发挥重要的自分泌和旁分泌作用 (Auernhammer & Melmed; Suman et al.)。人 LIF 可用于维持小鼠胚胎干细胞,但小鼠 LIF 无法与人类受体结合,因此导致小鼠 LIF 失活 (Dahéron et al.)。 LIF由CD4+细胞和活化的调节性T细胞产生,并促进Foxp3表达,同时抑制Th17谱系特异性基因(Metcalfe)。间充质基质细胞也分泌LIF,支持造血和免疫调节(Nasef et al.)。

(A) The biological activity of Mouse Recombinant LIF was tested by its ability to promote the proliferation of M1 cells. Cell proliferation was measured using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum. The EC50 in the above example is 2.7 pg/ml.
(B) 2 μg of Mouse Recombinant LIF was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Mouse Recombinant LIF has a predicted molecular mass of 19.9 kDa.
