白介素 12 (IL-12p70) 是由 p35 和 p40 亚基组成的异二聚体细胞因子。IL-12 由单核细胞、巨噬细胞、树突状细胞、中性粒细胞和 B 细胞在细菌产物和细胞因子,例如 IFN-γ 刺激下产生。IL-12 受体在 T 细胞、NK 细胞和树突状细胞上表达。结合后,IL-12 通过 JAK/STAT 信号通路启动信号传导,并刺激 NK 细胞、B 细胞和 T 细胞产生 IFN-γ (Watford et al.)。它还调节细胞因子的合成、T 细胞和 NK 细胞的增殖,并刺激 CD4+ 和 CD8+ T 细胞的分化 (Germann & Rüde)。缺乏 IL-12 的小鼠易受多种细胞内病原体的侵害,并且 IFN-γ 分泌、Th1 分化和 NK 细胞溶解活性受损;然而,Th2 发育和 IL-4 产生增强(Watford et al.)。

(A) The biological activity of Human Recombinant IL-12 was tested by its ability to produce IFN-gamma in NK cells co-stimulated with IL18. The EC50 is defined as the effective concentration of the growth factor at which IFN-gamma production is at 50% of maximum. The EC50 in the above example is less than 2 ng/mL.
(B) 2 μg of Human Recombinant IL-12 was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Human Recombinant IL-12 has a predicted molecular mass of 22.5 - 34.7 kDa.
