胶质细胞源性神经营养因子

胶质细胞源性神经营养因子(GDNF)是一种神经营养因子,是肿瘤生长因子 (TGF)- β超家族的成员。GDNF生长因子家族还包括neurturin、persephin和artemin,它们有7个保守的半胱氨酸残基,称为半胱氨酸结(Treanor et al.)。GDNF家族配体通过结合特异性GDNF家族受体-α (GFRα) 共受体发出信号,激活RET受体酪氨酸激酶(Durbec et al.)。目前已经鉴定出四种不同形式的GFRα共受体(GFRα 1-4),其中GDNF在与RET形成复合物之前特异性结合GFRα1 (Airaksinen & Saarma)。体内和体外研究表明,GDNF可促进中脑多巴胺能神经元的存活和形态分化,并增加其对高亲和力多巴胺的摄取(Granholm et al.; Lin et al.)。此外,GDNF还被证实对因退化毒素而死亡的多巴胺能神经元具有修复作用(Aoi et al.)。GDNF与人重组BDNF(脑源性神经营养因子;产品号 #78005)、BrainPhys™神经元培养基(产品号 #05790)和其他添加物一起可用于将人多能干细胞(hPSC)衍生的神经祖细胞分化为神经元(Bardy et al.)。本产品不含动物成分。

Data Figures

(A) The biological activity of Human Recombinant GDNF, ACF was tested by its ability to promote the proliferation of C6 cells. Cell proliferation was measured using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum. The EC50 in the example above is 1.84 μg/mL. (B) 1 μg of Human Recombinant GDNF, ACF was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining.
