概述

生长分化因子5 (GDF-5)是转化生长因子β (TGF-β)超家族骨形态发生蛋白(BMP)亚类的成员。它结合一个由BMPR1B和BMPR2组成的受体复合物,然后激活SMAD信号通路(Carreira等人;西藤等人;Osório等)。GDF-5在发育过程中促进间充质干细胞的软骨分化、骨骼形成和树突生长(Coleman等;Francis-West等人)。对大鼠的研究表明,GDF-5调节模式、神经发生、星形胶质发生和神经元规格(Gajavelli等人;Krieglstein等人;O’keeffe等人)。

(A) The biological activity of Human Recombinant GDF-5 was tested by its ability to induce alkaline phosphatase production in ATDC-5 cells. Alkaline phosphatase production was measured after 63 hours using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which alkaline phosphatase activity is at 50% of maximum. The EC50 in the above example is 0.017 μg/mL. (B) 1 μg of Human Recombinant GDF-5 was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining.
