M5/114.15.2(M5/114)抗体可特异性识别小鼠主要组织相容性复合物II类受体(MHC-II)上的胞外表位。MHC-II是一种异二聚跨膜糖蛋白,由一个α亚基与一个β亚基非共价结合而成。在小鼠中,MHC-II复合物被命名为I-A和I-E,分别包含Aα/Aβ和Eα/Eβ异源二聚体。MHC-II与CD3/T细胞受体(TCR)复合物以及CD4分子一起,介导抗原递呈细胞将外源抗原水解产生的肽递呈给CD4+ T(辅助性)细胞的关键功能,从而抑制或诱导对肽的免疫反应。M5/114.15.2抗体可识别I-Ab、I-Ad、I-Aq、I-Ed和I-Ek(但不包括I-Af、I-Ak或I-As) MHC II类同种抗原所共有的多态性决定因素,这些抗原在携带H-2b、d、p、q、r和u单倍型(但不包括H-2f或H-2s单倍型)的小鼠B细胞、树突状细胞、巨噬细胞、单核细胞和激活的T细胞上表达。研究发现,M5/114抗体抑制H-2b、d、q和u单倍型T细胞的I-A限制性反应(但不抑制H-2f、k或s单倍型),与NOD (H-2g7)小鼠细胞不发生反应。
