TRA-2-54/2J抗体与碱性磷酸酶（AP）的组织非特异性异构体反应，AP在未分化的多能干细胞，如人类和小鼠胚胎干细胞（ES），以及诱导多能干细胞（iPS）和胚胎癌（EC）细胞中高水平表达。AP的同工酶与肝、骨和肾细胞表达的同工酶相同。AP水平随着分化而降低，因此，TRA-2-54/2J抗体可用于监测多能干细胞的分化状态。TRA-2-54/2J抗体不与其他AP同工酶发生交叉反应，这些酶因翻译后修饰而不同，据报道，该抗体不识别来自牛、狗、山羊、豚鼠、仓鼠、小鼠、大鼠或羊的AP，也不识别恒河猴、狒狒和非洲绿猴等旧大陆猴子。
