小鼠单克隆IgG1抗体,抗人IFN-gamma
该小鼠单克隆抗体(克隆 1-D1K)能与人干扰素-γ(IFN-γ)反应。IFN-γ 是一种约 17 kDa 的蛋白质,由 T 细胞和 NK 细胞在有丝分裂和抗原刺激下产生,树突状细胞和巨噬细胞产生的量较少。IFN-γ 受 IL-12 和 IL-18 等细胞因子的控制,这些细胞因子是在感染时分泌的(Schroder et al.)。IFN-γ 是由相互连接的α-螺旋组成的同源二聚体(Thiel et al.),它与靶细胞上 IFN-γ 受体(IFNGR)的α亚基结合,导致受体交联并激活 JAK-STAT 信号传导(Kak et al.)。IFN-γ 可介导多种细胞效应,包括上调 I 类和 II 类抗原递呈途径、抗病毒和抗微生物机制、抗增殖和细胞凋亡作用、免疫调节和白细胞转运(Schroder et al.)。
