5C3抗体与CD40反应，CD40是一种45 - 50 kDa的I型跨膜糖蛋白，是肿瘤坏死因子受体（TNFR）超家族的成员。CD40在B细胞发育过程中发挥重要作用，也参与生发中心的B细胞选择过程。它通过与T细胞上表达的CD40配体CD154 （gp39）结合，促进T细胞- B细胞相互作用，从而产生共同刺激和免疫调节。在IL-4或其他刺激存在的情况下，CD40促进B细胞增殖，诱导免疫球蛋白（Ig）同型转换，并挽救B细胞免于从表面Ig诱导的凋亡。CD40表达于B细胞、滤泡树突状细胞、巨噬细胞、成纤维细胞、角质形成细胞、内皮细胞、基底上皮细胞、CD34+造血祖细胞和原发性癌的表面。它在浆细胞上表达水平很低，甚至根本不表达。5C3抗体已在体外功能试验中激活B细胞或诱导B细胞增殖。
