抗人CD3抗体(克隆SK7)可与人T细胞受体(TCR)/CD3复合物中分子量约为20 kDa的分化簇3 epsilon(CD3ε)亚基发生反应,该复合物在大多数(约95%)成熟T细胞和自然杀伤T细胞表面表达,在胸腺细胞上也有不同程度的表达。大多数T细胞肿瘤也表达CD3。CD3复合物由亚基 γ、δ、ε、η 和ζ组成,并与TCR非共价结合。它参与将抗原识别信号传导到T细胞的细胞质的过程中,并调节TCR在细胞表面的表达。TCR对T细胞的激活涉及CD3 亚基的细胞质尾部,其在结构上与1型跨膜蛋白和免疫球蛋白超家族成员相关。CD3亚基的突变与各种免疫缺陷病有关,包括严重联合免疫缺陷症(SCID)。
