总览

OKT3抗体可特异性识别人T细胞受体（TCR）/CD3复合物中约20 kDa的CD3ε亚基上的胞外构象表位，该表位在大多数（约95%）成熟T细胞和NKT细胞表面表达，在胸腺细胞上也有不同程度的表达，大多数T细胞肿瘤也表达CD3。CD3是由CD3γ、δ、ε、η和ζ亚基组成的二聚体，这些亚基在结构上属于免疫球蛋白超家族的1型跨膜蛋白。CD3与TCR非共价结合，介导抗原识别信号向T细胞胞内的转导，调控T细胞活化及TCR的膜表面表达。CD3基因突变与免疫系统疾病有关，包括重症联合免疫缺陷症（SCID）。 OKT3抗体可识别CD3ε亚基与CD3γ或CD3δ结合时表达的表位，该抗体的结合可诱导T细胞激活。由于其在体内具有免疫抑制特性，OKT3抗体已被证明可用于治疗同种异体移植排斥反应。克隆OKT3被广泛用于人T细胞表型分析，并已观察到其可阻断抗CD3抗体克隆SK7和UCHT1的结合。
