概述

白细胞介素21 (IL-21)是一种多效性细胞因子,由四个α-螺旋束组成,主要由自然杀伤T细胞(NKT)、T滤泡辅助细胞(Tfh)和Th17细胞产生(Spolski & Leonard 2008)。IL-21信号通过IL-21受体(IL-21R)的异源二聚体和il - 2rg编码的常见细胞因子受体γ链(Parrish-Novak et al.;Ozaki K et al. 2000),并利用JAK/STAT、MAPK和PI3K通路(Spolski & Leonard 2014)。IL-21已被证明在调节免疫球蛋白的产生和促炎Th17细胞群的分化中起关键作用(Ozaki等人,2002;Nurieva等人)。此外,IL-21特异性地维持CD8+ T细胞效应活性,并在慢性病毒感染期间提供CD4+ T细胞帮助的机制(Elsaesser等)。IL-21信号也被发现对NOD小鼠1型糖尿病的发展至关重要(Sutherland等人),并通过调节性B细胞控制T细胞自身免疫(Yoshizaki等人)。

(A) The biological activity of Human Recombinant IL-21 was tested by its ability to inhibit the proliferation of Mino cells. Cell proliferation was measured using a fluorometric assay method. The EC50 is defined as the effective concentration of the growth factor at which cell proliferation is at 50% of maximum. The EC50 in the example above is less than 0.5 ng/mL. (B) 2 μg of Human Recombinant IL-21 was resolved with SDS-PAGE under reducing (+) and non-reducing (-) conditions and visualized by Coomassie Blue staining. Human Recombinant IL-21 has a predicted molecular mass of 15.4 kDa.
