  • Isolate highly purified human CD56+ cells from freshly prepared buffy coat and leukoreduction system chamber (LRSC; also known as an LRS cone) samples by immunomagnetic positive selection, with the EasySep™ HLA Chimerism Buffy Coat CD56 Positive Selection Kit II. Widely used in published research for more than 20 years, EasySep™ combines the specificity of monoclonal antibodies with the simplicity of a column-free magnetic system.

    In this EasySep™ positive selection procedure, desired cells are labeled with antibody complexes recognizing CD56 and magnetic particles. Labeled cells are separated using an EasySep™ magnet and by simply pouring or pipetting off the unwanted cells. The cells of interest remain in the tube. Following magnetic cell isolation, the desired CD56+ cells are ready for downstream applications such as flow cytometry, culture, or DNA/RNA extraction for lineage-specific chimerism analysis. The CD56 antigen is expressed on NK cells and NKT cells.

    This product replaces the EasySep™ Human Buffy Coat CD56 Positive Selection Kit (Catalog #18085) and EasySep™ HLA Buffy Coat CD56 Positive Selection Kit (Catalog #18085HLA), for even faster cell isolations.

    Starting with freshly prepared buffy coat, the CD56+ cell content of the isolated fraction typically ranges from 95.8 - 99.5% (gated on CD45+ using “The Big Easy” EasySep™ Magnet). In the above example, the purities of the start and final isolated fractions are 8.5% and 98.4%, respectively. NOTE: RBCs were removed from the start sample by lysis prior to flow cytometry.
