DNA甲基转移酶抑制剂
概述
用地西他滨抑制DNA合成，地西他滨是一种结合DNA甲基转移酶的低甲基化剂。地西他滨是一种胞嘧啶核苷类似物，在DNA复制过程中被纳入DNA链。当DNA甲基转移酶与DNA结合并将甲基化复制到子链上时，地西他滨与DNA甲基转移酶结合并阻止其继续DNA复制（Kantarjian等人）。
地西他滨用于：
癌症研究
·抑制A375、SKMEL1、SKMEL3、SKMEL28、MeWo和B16人类皮肤黑色素瘤细胞系的增殖（Alcazar等）。
·通过增加活性氧的产生诱导人白血病细胞系U937和HL60细胞凋亡（Shin等）。
·诱导AGS胃癌和A549肺癌细胞G2/M期细胞周期阻滞（Shin等）。
·降低Kasumi-1AML细胞系的增殖能力，并略微增加细胞凋亡（Flotho等人）。
·在口腔鳞状细胞癌和肝癌细胞系中通过上调p164a诱导细胞衰老(Suh等；Timmermann等人)。
·诱导HepG2和Hep3B细胞系细胞衰老和p53依赖性肿瘤细胞衰老，以及大量DNA双链断裂（Venturelli et al.）。
