12G5 抗体可识别 CD184(又称 CXCR4 或 fusin),该蛋白是一种约 45 kDa 的七次跨膜 G 蛋白偶联受体,广泛表达于多种细胞表面,包括 T 细胞、B 细胞、树突状细胞、单核细胞、巨噬细胞、粒细胞、血小板、内皮细胞、上皮细胞、星形胶质细胞和神经元。CD184 至少有两种亚型,由可变剪接的 mRNA 转录产生。其配体包括 CXC 趋化因子 SDF-1 和泛素。CD184 在趋化反应、B 细胞淋巴生成、髓系细胞生成、心脏发育、血管生成及神经引导等过程中发挥重要功能。它还作为某些 HIV-1 T 细胞嗜性分离株感染的共受体,以及 HIV-2 在不依赖 CD4 的情况下感染细胞的替代受体,而这些功能可被 12G5 抗体结合后阻断。该抗体还可部分抑制 SDF-1 诱导的趋化作用和钙离子内流。在分化研究中,CD184 常与其他标志物(如 CD117)联合使用,以监测定型内胚层的出现。
