概述

本试剂盒可从新鲜外周血单个核细胞（PBMCs）或洗涤后的白细胞单采术样本中分选出人CD62L+CD45RO+和CD62L-CD45RO+记忆CD8+ T细胞。首先，通过使用抗体复合物和EasySep™ Releasable RapidSpheres™的免疫磁珠正选无柱分选CD62L+细胞。随后，结合在EasySep™分离的CD62L+细胞上的磁珠被解离，CD45RA+和非CD8+ T细胞被抗体复合物和EasySep™ Dextran RapidSpheres™识别并去除。最终分选后的细胞中含有高纯度的CD62L+CD45RO+中央记忆CD8+ T细胞，可立即用于下游应用。效应记忆CD8+ T细胞可从CD62L-组分中继续分选获得。

Starting with fresh PBMCs, the central memory CD8+ T cell content (CD3+CD8+CD45RO+CD62L+) of the isolated fraction is typically 86.8 ± 8.4% (mean ± SD using the purple EasySep™ magnet). The effector memory content (CD3+CD8+CD45RO+CD62L-) of the isolated fraction is typically 88.7 ± 6.5% (mean ± SD).
NOTE: RBCs were removed from the start sample by lysis prior to flow cytometry.
